Imaging based approaches to localization of proteins at the subcellular level provide spatial information in situ from single cells and can effectively address the issue of proteins that localize to multiple organelles. Endogenous proteins are best visualized by the use of specific antibodies and fluorophores fused to secondary antibodies in fixed cells. Another common technique is target proteins fused to a fluorescent protein thus enabling temporal studies of the protein’s distribution in its natural environment.
