Introduction
The PerkinElmer IVIS Lumina X5 combines high-throughput in vivo bioluminescence and fluorescence imaging with high-resolution 2D X-ray.
The IVIS Lumina X5 instrument provides 20 x 20 cm field of view, for imaging up to 5 animals at a time, ex-vivo specimens and cell culture plates with bioluminescence and fluorescence. A large, independently deployed scintillator enables X-ray acquisitions of large rodents, up to 600 grams or 5 mice.
The IVIS Lumina X5 is equipped with 26 filters (19 excitation filters and 7 emission filters) to image fluorescent sources that emit from green to near-infrared. The X-ray module employs automated image integration to overlay with bioluminescence, fluorescence and photography.
The system is equipped with an isoflurane anesthesia system, low-autofluorescence optics and a cooled CCD camera (‑90°C) for a maximum resolution of 50µm.

Analysis
The IVIS Lumina X5 Living Image software (Living Image 4.5) allows absolute quantification (photons/sec/cmsq/sr) of bioluminescence and fluorescence signal, for in vivo, ex vivo and in vitro studies.
The software supports accurate autofluorescence removal, unmixing, fluorophore quantitation, automatic ROI creation, background subtraction and scaling.
The software is available both at the Lumina X5 station and at the BCF image analysis unit on our Computer in-vivo analysis.
